    摘要: The oriental fruit fly, Bactrocera dorsalis (Diptera: Tephritidae) is a destructive horticultural pest which causes considerable economic losses every year. A collection of microorganisms live within the B. dorsalis gut, and they are involved in its development, physiology, and behavior. However, knowledge regarding the composition and function of the gut mycobiota in B. dorsalis are still limited. Here, we comprehensively characterized the gut mycobiota in B. dorsalis across different developmental stages. High-throughput sequencing results showed a significant difference in fungal species abundance and diversity among different developmental stages of B. dorsalis. Quantitative polymerase chain reaction and culture-dependent methods showed that yeast species was the dominant group in the larval stage. We isolated 13 strains of yeast from the larval gut, and found that GF (germ-free) larvae mono-associated with strain Hanseniaspora uvarum developed faster than those mono-associated with other tested fungal strains. Supplementing the larval diet with H. uvarum fully rescued B. dorsalis development, shortened the larval developmental time, and increased adult wing lengths, as well as the body sizes and weights of both pupae and adults. Thus, our study highlights the close interactions between gut fungi, especially H. uvarum, and B. dorsalis. These findings can be applied to the sterile insect technique program to promote host development during mass insect rearing.

    摘要: Toll-like receptors (TLRs) are important initiators of innate immune responses that target host-pathogen interactions. However, further research into the molecular characteristics of TLRs in wild populations is required, as well as how TLRs genetically influenced pathogen infections in the brown rat (Rattus norvegicus). Here, we explored the genetic characterization and evolution of 2 sensing nucleic acid TLRs (TLR7 and TLR8) and 2 sensing non-nucleic acid TLRs (TLR2 and TLR4) in the wild brown rat, and assessed their associations with 2 RNA viruses (Seoul hantavirus and rat hepatitis E virus (HEV)) and 2 bacteria (Leptospira and Bartonella). In these 4 TLRs, we discovered a total of 16 variants. Furthermore, TLR8 had high genetic diversity among 7 variants, while TLR2 had low genetic diversity with only 1 variant. According to selective pressure analyses, TLR4, TLR7, and TLR8 genes evolved under purifying selection. Interestingly, significant associations were found between 3 TLR8 variants and HEV infection, as well as 1 TLR2 variant and Bartonella infection. Overall, our findings provided a glimpse into the genetic characterization of TLRs in the brown rat, and further demonstrated that TLR2 and TLR8 genetic variations were related to Bartonella and HEV infection, respectively. Especially, TLR8 may be a good candidate immune gene for future research on molecular ecology and functional adaptation in wild populations.

    摘要: Identifying the mechanism for sex determination in amphibians is challenging. Very little is known about sex determination mechanisms of Rana dybowskii, a species of importance to evolutionary and conservation biology. We screened for sex-linked molecular markers in R. dybowskii in China using target region amplification polymorphism with 2 fixed primers against the sequences of Dmrt1. We found 2 male-linked molecular markers in R. dybowskii, which were 222 bp and 261 bp long. The detection rates of 222 bp marker in males form Xinglong, Huadian, and Dandong were 93.79%, 69.64%, and 13.64%, respectively, while the rate in females from Huadian was 27.50%. Besides, the detection rates of 261 bp marker in the above 3 regions were only observed in males at the rate of 93.79%, 87.50%, and 32.73%, respectively. The inheritance patterns of sex-linked molecular markers showed that the 2 sex-linked molecular markers were heterozygous. Compared to the XY-male parent, progeny from XX-pseudo-male parent possessed lower sex reversal ratio at the same rearing temperature, and the proportion of female froglets from an XX-pseudo-male parent was more than 95% at low rearing temperature (15 degrees C). Our findings suggest that R. dybowskii displays male heterogamety, and the 2 sex-linked molecular markers may have a guiding significance for the protection and utilization of R. dybowskii.

    摘要: The effect of combined levonorgestrel (P) and quinestrol (E) on the fertility of striped field mouse (Apodemus agrarius) has not been evaluated. We performed a series of experiments in both the laboratory and field to assess the effect of P and/or E on the fertility of A. agrarius. In the laboratory, to test the time-dependent anti-fertility effects of P and E, as well as their mixtures, 90 male striped field mice were randomly assigned to 6 treatment groups (n = 60), and a control group (n = 30). Mice in 3 treatment groups were administered 1 of the 3 compounds (1 mg.kg(-1) [body weight] EP-1, 0.34 mg.kg(-1) E, 0.66 mg.kg(-1) P) for 3 successive days (another half for 7 successive days) via oral gavage; mice were then sacrificed 15 and 45 days after initiating the gavage treatment. Our findings indicated that E and EP-1 treatment, but not P or control treatment, significantly decreased the sperm count in the caudal epididymis, as well as the weight of the testes, epididymides, and seminal vesicles. Additionally, fertile female mice mated with E- and EP-1-treated males produced smaller pups. These data indicate that E and EP-1 can induce infertility in male A. agrarius. In the field, the population density of A. agrarius was significantly influenced by EP-1, and the rodent density in the treatment group was lower than that in the control group. Overall, our results indicate that EP-1 is an effective contraceptive in A. agrarius, a dominant rodent species in the farmland.

    摘要: The overall infection rate of Pentatrichomonas hominis in Siberian tigers in northeast China is 31.3%. All the P. hominis identified in Siberian tigers belonged to genotype CC1.
